Anacor Pharmaceuticals Inc announced that it has priced an underwritten public offering of 3,129,890 shares of its common stock at a price to the public of $6.39 per share for gross proceeds of $20.0 million. Anacor has also granted the underwriters a 30-day option to purchase up to 15 percent of the shares of common stock offered in the public offering to cover over-allotments. Anacor Pharmaceuticals Inc Announces Positive Results From First Of Two Phase 3 Trials Of Tavaborole For Onychomycosis
Anacor Pharmaceuticals Inc announced positive preliminary results from first of two Phase 3 trials of tavaborole, topical anti-fungal for onychomycosis, a fungal infection of nail and nail bed that affects approximately 35 million people in United States. Tavaborole achieved a high degree of statistical significance on all primary and secondary endpoints. In this first Phase 3 study (known as Study 301), 6.5% of patients treated with tavaborole met primary endpoint of complete cure vs. 0.5% of patients treated with vehicle (p=0.001) at week 52. Complete cure is a composite endpoint that requires both a mycological cure and a completely clear nail. Study 301 enrolled 594 patients in United States and Mexico with distal subungual onychomycosis, randomized two-to-one to receive either tavaborole, 5% solution, or vehicle. Eligible patients were at least 18 years of age (with no upper age limit) with a clinical diagnosis of distal subungual onychomycosis involving 20% - 60% of the total area of target great toenail, at least 3mm of clear nail from proximal nail fold to the proximal visible mycotic border and positive mycology. Patients were instructed to apply tavaborole solution or vehicle to the target great toenail once daily for 48 weeks. Overall, tavaborole was safe and well-tolerated across study subjects. There were no serious adverse events related to study drug. The rate of discontinuations as a result of adverse events was low (2.8% for tavaborole and 1.6% for vehicle). Full Article

Anacor Pharmaceuticals Inc's Partner GlaxoSmithKline plc Discontinues Clinical Development Of GSK2251052
Anacor Pharmaceuticals Inc announced that collaboration partner GlaxoSmithKline plc (GSK) has discontinued clinical development of GSK2251052. As previously disclosed, GSK halted the recent clinical trials of GSK2251052 due to the identification of microbiological findings of resistance in a small number of patients in the Phase 2b trial for the treatment of complicated urinary tract infections (cUTI). Since then, GSK has conducted additional pre-clinical research and, after assessing various options, has elected to discontinue further development of GSK2251052 and return all rights to GSK2251052 to Anacor. Anacor and GSK are currently working together to transition GSK2251052 back to Anacor. Anacor licensed GSK2251052 to GSK in July 2010 under the companies' ongoing research and development collaboration. GSK is continuing to fund Anacor’s research activities in tuberculosis as announced in September 2011 under the companies’ amended R&D collaboration. Full Article

Anacor Pharmaceuticals Inc announced positive preliminary results from two safety studies of AN2728 - a maximal use systemic exposure (MUSE) study in psoriatic patients and a local tolerability study. The results of these two studies demonstrate that AN2728 Ointment, 2% appears to be safe and well-tolerated when applied to very large body surface areas and that it is well-tolerated when applied to areas of sensitive skin. Anacor Pharmaceuticals Inc Announces Partner GSK Voluntarily Paused Enrollment Of Ongoing GSK ‘052 Phase 1 And 2 Clinical Trials
Anacor Pharmaceuticals Inc announced that its partner GlaxoSmithKline (GSK) has voluntarily paused enrollment in its current clinical trials of GSK2251052 (GSK ‘052) due to a recently identified microbiological finding in a small number of patients in the Phase 2b trial of GSK ‘052 for the treatment of complicated urinary tract infections (cUTI). GSK is in the process of obtaining additional information in order to better understand the data, but will not resume the trials until it has concluded its investigation. Upon completion of the review, GSK will determine next steps regarding the future development of GSK ‘052. In addition to the Phase 2b study in cUTI, GSK ‘052 is currently being evaluated in a Phase 2b study in complicated intra-abdominal infections (cIAI) and two Phase 1 studies in healthy volunteers. While the microbiological finding seen in the cUTI study is not related to the safety of GSK ‘052, the potential to negatively impact efficacy led GSK to voluntarily suspend enrollment in all four studies. Anacor Pharmaceuticals Inc Announces Positive Preliminary Results From Phase 2a Trial Of AN2728 And AN2898 In Atopic Dermatitis
Anacor Pharmaceuticals Inc announced positive preliminary results from a Phase 2a trial of its two boron-based phosphodiesterase-4 (PDE-4) inhibitors, AN2728 and AN2898, in mild-to-moderate atopic dermatitis, a chronic rash characterized by inflammation and itching. The primary endpoint for both compounds was successfully achieved after 28 days of twice-daily treatment, with 64% of AN2728-treated lesions showing improvement in Atopic Dermatitis Severity Index (ADSI) score versus 24% for vehicle (P = 0.05) and 71% of AN2898-treated lesions showing improvement in ADSI score versus 14% for vehicle (P = 0.01).
